Optimization of Determination Method of Total Flavonoids from Eucommia Ulmoides Oliv. Leaves

摘要 目的:优化杜仲叶总黄酮含量测定方法。方法:将大孔树脂吸附和Al(NO3)3络合法相结合,测定杜仲叶中总黄酮的含量,并对加入3种化学试剂后的显色静置时间进行优化。结果:优化的测定方法在减少干扰物质的同时,使3种显色剂的总显色时间由原先的27min减少到18min;在稳定的显色时间内,测定结果的平均加样回收率为98.52%,RSD1.12%(n=3)。结论:优化的方法具有分析速度快,准确度高,重现性好,精密度高,显色稳定性好等优点,是一种测定杜仲叶总黄酮含量的理想方法,并可为其他原料中总黄酮含量的测定提供参考依据。 Objective:To optimize the measurement of the total flavonoids in Eucommia ulmoides Oliv. Leaves. Method: The total content of the flavonoids were determined by the Al(NO3)3 colorimetry combined with the macroporous resin adsorption method and the standing time after adding chromogenie solutions was optimized too. Results: The inter- fering substances were reduced and the total standing time after adding three chromogenie solutions was decreased from 27 rain to 18 rain by use of this optimal method. Within the stable reaction time, the average value of the recovery for the determination was 98.52% with 1.12% of RSD(n=3). Conclusion: The optimal method was quick, accurate, repro- ducible and stable. It was an ideal method to determine total flavonoids in Eucommia ulmoides Oliv. Leaves. Futhermore, it could be looked as a reference method for the determination of total flavonoids in other materials.
