TNF-α对人软骨细胞中ChM-Ⅰ表达的影响

摘要 目的研究肿瘤坏死因子-α(tumor neerosis factor alpha,TNF-α)对人软骨细胞中软骨调节素-Ⅰ(chondro-modulin-Ⅰ,ChM-Ⅰ)表达的影响。方法用浓度为5ng/mL、10ng/mL和20ng/mL的TNF-α处理细胞24h、48h和72h,荧光实时定量聚合酶链式反应(real-time quantitative polymerase chain reaction,RT-qPCR)和蛋白免疫印迹(western blot)方法检测细胞中ChM-Ⅰ在mRNA和蛋白水平的表达量。结果 TNF-α处理后,各浓度组在各时间点均可下调ChM-Ⅰ的表达(P<0.05),且随浓度和时间的增加,下调作用增强,以20ng/mL处理72h时下调作用最强,下调37%;western blot结果与RT-qPCR相一致。结论肿瘤坏死因子-α可下调人软骨细胞中ChM-Ⅰ的表达。 Objective To investigate the effect of Chondromodulin- I (ChM- I ) expression in human cartilage cells by tumor necrosis factor alpha (TNFα). Methods TNF-α was used to stimuli human cartilage cells for 24 h,48 h and 72 h in the concentration of 5 ng/mL,10 ng/mL,20 ng/mL,and the expression of ChM- I in the cells were measured in the levels of mRNA and protein by real-time quantitative polymerase chain reaction (RT-qPCR) and western blot. Results After TNF-α stimulating,the expression of ChM- I was decrease in the levels of mRNA and protein. And the effect was show to be time and dose dependent. Conclusion TNF-α can down-regulate the expres- sion of ChM- I in human cartilage cells.
