风湿Ⅱ号胶囊对佐剂性关节炎大鼠血清肿瘤坏死因子-α和白细胞介素的影响

Effects of Fengshi Ⅱ Capsule on Tumor Necrosis Factor-α and Interleukin in Adjuvant Arthritis Rats

目的观察风湿Ⅱ号胶囊对佐剂性关节炎(AA)大鼠血清中肿瘤坏死因子-α(TNF-α)与白细胞介素(IL)-4、IL-10的影响,并探讨其作用机制。方法除空白组外,采用弗氏完全佐剂注射于大鼠右后足跖皮内制作AA大鼠模型,并随机分为模型组、阳性药(雷公藤多甙片)组及中药高、中、低剂量组。中药高、中、低剂量组分别以风湿Ⅱ号胶囊药液0.648、0.324、0.162 g/kg灌胃,阳性药组用雷公藤多甙片6.0 mg/kg灌胃,空白对照组及模型组给予等容积蒸馏水,每日1次。用药28 d后,观察治疗前后各组大鼠的一般情况、体质量变化、关节炎指数,并测定各组大鼠血清中TNF-α、IL-4、IL-10水平。结果与空白组比较,模型组大鼠体质量增长缓慢,血清中TNF-α显著升高,而IL-4及IL-10则明显下降(P<0.01);与模型组比较,中药各组及阳性药组大鼠的体质量增长快,关节炎指数明显降低,且均可降低TNF-α含量,升高IL-4、IL-10水平(P<0.05或P<0.01);与阳性药组比较,中药高剂量组能显著增加AA大鼠的体质量(P<0.05),降低关节炎指数(P<0.01),降低血清中TNF-α水平(P<0.01),中药高、中剂量组则能显著升高IL-4、IL-10水平(P<0.01)。结论风湿Ⅱ号胶囊通过降低血清中TNF-α含量,升高IL-4、IL-10水平,恢复类风湿性关节炎的细胞因子间的平衡,从而达到防治类风湿性关节炎的作用。

Objective To observe the effects of Fengshi Ⅱ capsule on TNF-α,IL-4 and IL-10 in adjuvant arthritis（AA） rats,and explore its mechanism.Methods The AA models were built by Freund＇s complete adjuvant.The rats were randomly divided into normal group,model group,positive control（tripterygium polyglucoside） group,and low-,medium-and high-dose herbs groups.The herbs groups were treated for 28 days with Fengshi Ⅱ capsule 0.648,0.324,0.162 g/kg respectively,the positive control group with tripterygium polyglucoside at a dose of 6.0 mg/kg,normal group and model group with same volume of distilled water.The health condition,body weight and arthritis index were observed,and TNF-α,IL-4 and IL-10 in the serum were determined.Results In the model group,the level of TNF-α increased compared with the normal group（P 0.01）,but the levels of IL-4 and IL-10 significantly descended（P 0.01）.After treatment with tripterygium polyglucoside and herbs,the level of TNF-α and arthritis index were lower than that in model group and the levels of IL-4 and IL-10 were higher than that in model group（P 0.05 or P 0.01）.The level of TNF-α and arthritis index of the high-dose herbs group significantly descended compared with the positive control group,but the levels of IL-4 and IL-10 of the medium-dose and high-dose herbs groups increased（P 0.01）.Conclusion Fengshi Ⅱ capsule could treat rheumatoid arthritis by decreasing TNF-α,increasing IL-4 and IL-10 to adjusting the levels of cytokines.
