摘要
以T7RNA聚合酶或RNA聚合酶Ⅰ/Ⅱ为基础的病毒反向遗传操作系统在设计构建和应用上由繁到简不断改进。本文分别以新城疫病毒、狂犬病病毒和禽流感病毒等负链RNA病毒为例,综述了近十年来病毒反向遗传操作中的构建策略创新及其在分子病毒学和反向疫苗学的应用,旨在为更好地利用该技术提供参考。
The reverse genetic system based on T7 RNA polymerase or RNA polymerase Ⅰ/Ⅱ was innovated from complex to simple in principle for its construction and application. The negative strand RNA viruses, newcastle disease virns, rabies virus and avian influenza virus were reviewed to summarize the technological innovation on reverse genetics and ils applications in molecular virology and reverse vaceinology in recent 10 years. This infor marion will provide useful references for a better understanding of the reverse genetics technology in this field.
