解毒化瘀颗粒对慢性重型肝炎患者外周血单个核细胞Toll样受体4表达调控的研究 被引量:6

摘要 目的:明确解毒化瘀颗粒对慢性重型肝炎患者外周血单个核细胞(PBMC)TLR(Toll样受体)4表达的调控作用,初步阐明解毒化瘀颗粒缓解内毒素血症的作用机制。方法:将慢性重型肝炎患者110例,随机分为对照组55例,治疗组55例,采用流式细胞技术检测两组患者治疗第2周、4周外周血PBMC TLR4表达情况。结果:两组患者外周血PBMC TLR4均呈高表达状态。治疗第2周,对照组测TLR4(%)表达为51.48±11.35,治疗组为47.56±12.14,差异无统计学意义(P>0.05)。治疗第4周,治疗组患者外周血PBMC TLR4表达明显低于对照组,其中对照组为43.97±9.56,治疗组为37.61±8.59,差异有统计学意义(P<0.05)。结论:在西医综合治疗的基础上,加用解毒化瘀颗粒能显著降低慢性重型肝炎患者外周血PBMC TLR4表达,改善内毒素血症。表明解毒化瘀颗粒对LPS(脂多糖,为细胞内毒素化学成分)/TLR4信号传导通路有调控作用。 Objective:To study the regulation effect of Jieduhuayu granule on TLR4 expression in peripheral blood mononuclear cell of patient with chronic severe hepatitis, preliminarily clarify the mechanism of Jieduhuayu granuld for alleviating endotoxemia. Methods:One hundred and ten patients with chronic severe hepatitis were randomly divided into control group and treatment group equally, using flow cytometry to detect TLR4 expression in peripheral blood mononuclear cells at the second and fourth week. Results: The treatment group and control group's TLR4 into a high expression on week two, the expression of TLR4 (%) was 51.48 ± 11.35 in control group, 47.56±12.14 in treatment group, the difference was not statistically significant (P〉 0.05). On the fourth week, the expression of TLR4 in treatment group was significantly lower than the control group, the control group was 43.97 ± 9.56, and the treatment group was 37.61 ± 8.59, the difference is statistically significant (P〈0.05). Conclusion: Based on western medicine treatment, Jieduhuayu granule particles can significantly reduce peripheral monocytes TLR4 expression, improve endotoxemia. Show that Jieduhuayu granule can regulate LPS/TLR4 pathway.
