悬钩子属野生种及品种亲缘关系的ISSR分析 被引量:6

摘要 利用ISSR分子标记方法,对39份悬钩子属(Rubus L.)野生种或变种及17份栽培品种进行分析,探讨这56份材料间亲缘关系的远近。通过试验从35个引物中筛选出12个能扩增出清晰带型并具多态性的引物,共扩增出166条DNA片段,分子量在250~2300 bp之间,平均每条引物扩增出13.83条DNA片段,多态性比率为96.33%,然后采用NTSYS-pc软件和UPGMA法构建了系统树。聚类分析结果将野生种分成5大类群,其中15份栽培品种包含在内,而有2份栽培品种(酷它它和宝森)另成一类,区别于其他54份材料。试验认为ISSR分子标记技术适合作悬钩子属植物组间亲缘关系的分析。 Genetic relationship of 39 Rubus L. wild species or varieties and 17 euhivars were studied using inter-simple sequence repeat( IS-SR) marker. 12 primers, which were screened out from 35 primers, could get clear and polymorphic fragments. A total of 166 DNA frag-ments ranging from 259 bp to 2300 bp were got through PCR by these 12 primers. The average number of DNA fragments produced by each primer was 13.83. The polymorphic rate was 96.33 %. A dendregram of showing genetic relationships was constructed by using the NTSYS- pc software and UPGMA. The 39 wild species and 15 euhivars were divided into 5 groups by clustering analysis, and other 2 cultivars( Kota-ta and Boysen) belonged to another groups and differented from the 54 a materials. The results showed that ISSR molecular marker technique was suitable for analyzing the Rubus interspeeifie relationship.
