摘要
目的:应用近红外光谱法建立一种逍遥丸(浓缩丸)中芍药苷含量的快速测定方法。方法:利用高效液相色谱法测定样品中芍药苷含量,并以其为参考值,运用偏最小二乘法(PLS)建立芍药苷含量与近红外光谱之间的多元校正模型,对未知样品进行含量预测。结果:所建芍药苷定量分析模型的相关系数(R2)、内部交叉验证均方差(RMSECV)、校正均方差(RMSEC)分别为0.997 75、0.350 93、0.048 8;经内部验证,模型的预测均方差(RMSEP)和平均回收率分别为0.082 8和100.08%。结论:近红外光谱法操作简便,无污染,结果准确可靠,可用于逍遥丸(浓缩丸)中芍药苷含量的快速测定。
Objective:To establish a rapid analytical method for Paeoniflorin in Xiaoyao Pills (condensed) by Near-infrared Spectrosco- py. Methods:The near-infrared spectral data of 106 samples was collected by Nicolet 6700 NIR speetroradiometer, and the data of Pae- oniflorin content were obtained by HPLC. Then, the multivariate calibration model for Paeoniflorin was established by patrical least square (PLS) with HPLC analysis values as reference, and predicting the Paeoniflorin content of unknown samples. Results:the correla- tion coefficients (R2) of the quantitative calibration model for Paeoniflorin was 0. 997 75, the root-mean-square error of cross-validation (RMSECV) was 0. 350 93, the root-mean-square error of calibration (RMSEC)was 0. 048 8;the root-mean-square error of prediction (RMSEP) and the average rate of recovery were 0.082 8 and 100.08%. Conclusion : The method is simple, non-pollnted, accurate, and could be applied for the fast determination of Paeoniflorin in Xiaoyao Pills (condensed).
