青风藤醇提液对关节炎大鼠关节骨破坏的影响 被引量:15

The Effect of Alcohol Extracts from Caulis Sinomenii on Articular Bone Destruction of Rats with Arthritis

摘要 目的观察青风藤醇提液对胶原诱导关节炎(CIA)大鼠关节破坏的影响,初步研究其作用机制。方法雌性SD大鼠32只,随机分为对照组、模型组和青风藤治疗组,模型组和青风藤治疗组制备CIA模型。青风藤治疗组灌胃青风藤醇提液,模型组与对照组灌胃等量0.9%氯化钠溶液。每3 d行关节评分1次,ELISA法检测血清白细胞介素-17(IL-17)、核因子κB(NF-κB)配位体受体活化剂(RANKL)、可溶性蛋白骨保护素(OPG),观察大鼠后足病理组织切片。结果关节评分模型组显著高于青风藤治疗组(P<0.01);3组大鼠IL-17表达量均差异有统计学意义(P<0.05),IL-17水平青风藤治疗组较模型组低(P<0.05),较对照组高(P<0.01)。3组大鼠血清RANKL、OPG水平及RANKL/OPG值均差异有统计学意义(P<0.01),大鼠血清RANKL水平青风藤治疗组较模型组低(P<0.05),OPG水平青风藤治疗组较模型组高(P<0.05)。RANKL/OPG青风藤治疗组低于模型组,高于对照组;病理结果显示,大鼠关节骨破坏程度青风藤治疗组明显低于模型组。结论青风藤醇提取液能降低CIA大鼠血清IL-17、RANKL水平,升高OPG水平,降低RANKL/OPG,抑制破骨细胞生成,发挥抑制关节骨破坏作用。 Objective To investigate the effect of alcohol extracts from Caulis sinomenii on articular bone destruction of rats with collagen-induced arthritis ( CIA), and preliminarily explore its mechanism of action. Methods The total of 32 female SD rats were randomly divided into the control and model groups, and the latter and the latter was randomly derided into model control and Caulis sinomenii groups. The Caulis sinomenii group was treated with alcohol extracts from Caulis sinomenii,while the model and control groups were treated with equal volume of saline solution. The feat of the articular score was observed every three days ;the serum levels of IL-17, RANKL and OPG were test by ELISA. The dead rats" feet were taken for histopathologic slice and HE staining to detect the bone destruction. Results The articular score of model group was significantly higher than that of the Caulis sinomenii group ( P〈0.01 ) ; IL-17 level of three groups showed a statisticant difference ( P〈0.05 ), IL-17 of the Caulis sinomenii group was remarkablly lower than that of the model group (P〈0.05) , but higher than that of the control group( P〈 0.01 ). The levels of RANKL, OPG and RANKL/OPG ratio of the three groups also showed obviously difference ( P〈0.01 ). The level of RANKL of Caulis sinomenii group was lower than that of the model group( P〈0.05 ) and the OPG significantly higher than that of the model group (P〈0.05). The RANKIVOPG ratio of the Caulis sinomenii group was fewer than that of the model group ,but more than that of the model group;the HE staining results showed that the extent of article bone destruction of Caulis sinomenii group was less severe than that of the model group. Conclusion Caulis sinomenii can inhibit articular bone destruction CIA rats" by reducing the peripheral by reducing the peripheral serum of RANKL and IL-17, elevating OPG level, down-regulating RANKL/OPG ratio and thus inhibiting intra-articular osteoclastogenesis.
