BDNF对哮喘小鼠C7~T5节段脊髓后角p-CREB、CREB表达的上调作用

摘要 目的探讨哮喘发病中脑源性神经营养因子(BDNF)对哮喘小鼠C7~T5节段脊髓后角内cAMP反应元件结合蛋白(p-CREB、CREB)表达的调节作用。方法 BALB/c小鼠30只,按随机数字表法均分为正常对照组、哮喘组、Anti-BDNF抗体组,利用AniRes2005肺功能仪测小鼠气道阻力、免疫荧光方法检测各组小鼠C7~T5节段脊髓后角内p-CREB、CREB的表达。结果哮喘组小鼠吸气阻力和呼气阻力明显高于正常组(P<0.01);与哮喘组相比,Anti-BDNF抗体组吸气阻力和呼气阻力则明显降低;免疫荧光结果显示哮喘组C7~T5节段脊髓后角内p-CREB、CREB阳性产物的平均光密度值(MOD)显著低于正常对照组(P<0.01);而anti-BDNF抗体组与哮喘组相比,p-CREB、CREB阳性反应产物的MOD值明显升高(P<0.01)。结论 BDNF能上调哮喘小鼠C7~T5节段脊髓后角内p-CREB、CREB的表达。 Objective To study the regulatory effect of brain-derived neurotrophic factor (BDNF)on the expression of cAMP re- sponse element binding protein( p-CREB, CREB)in the spinal dorsal horn of the asthmatic mice. Methods 30 BALB/c mice were randomly divided into normal control, asthmatic, anti-BDNF groups on average. The airway resistance of mice was measured with AniRes 2005 pulmonary function meter. The expressions of p-CREB, CREB were observed by immunofluorescence in the C7 - T5 spinal dorsal horn of all the groups. Results Inspiratory resistance and expiratory resistance in the asthmatic mice were obviously higher than those in normal control mice; compared with the asthmatic mice, inspiratory resistance and expiratory resistance were decreased significantly in anti-BDNF group. The mean optic density(MOD) of p-CREB, CREB positive product in the C7-T5 spinal dorsal horn were increased significantly in the asth- matic mice than those in normal control by means of immunofluorescence (P 〈0. 01 ) ; but decreased in the anti-BDNF group than those in the asthmatic mice (P 〈0. 01 ). Conclusions BDNF might be upregulate the expressions of p-CREB, CREB in the C7-T5 spinal dorsal born of the asthmatic mice.
