Aβ_(1~42)寡聚体与纤维体的制备及鉴定 被引量:6

摘要 目的探讨Aβ1~42寡聚体和纤维体的制备及其鉴定方法,进而比较两者的细胞毒性。方法设定不同的环境条件将Aβ1~42单体分别聚集成寡聚体和纤维体两种状态,利用原子力显微镜观察Aβ1~42的不同聚集状态。CCK-8法比较Aβ1~42寡聚体与纤维体对BV-2细胞存活率的影响。结果原子力显微镜下观察Aβ1~42寡聚体呈球状或椭圆球状颗粒,高度为5 nm左右,Aβ1~42纤维体在镜下呈现长条纤维状聚集,高度约为3 nm左右,长度>1 000 nm。CCK-8法显示Aβ1~42寡聚体和纤维体均对BV-2细胞具有损伤作用,并成剂量-效应关系。在同一浓度水平条件下,Aβ1~42寡聚体组的BV-2细胞存活率显著低于纤维体组(P值<0.05)。结论原子力显微镜可直观地观察到Aβ1~42的不同聚集状态,Aβ1~42寡聚体比纤维体更具细胞毒性。
