摘要
目的探讨阿司匹林对宫颈癌Hela细胞增殖的影响。方法体外培养Hela细胞,分别以不同浓度阿司匹林干预。应用Giemsa染色观察细胞形态学的改变;琼脂糖电泳法观察凋亡细胞DNA Ladder现象;采用MTT法测定细胞增殖抑制率。结果 Giemsa染色后可见阿司匹林组细胞体积缩小,凋亡小体形成,且随着浓度增加,凋亡率也增加;不同浓度阿司匹林作用Hela细胞48 h后,提取DNA电泳,可见明显的DNA裂解带;MTT结果显示,在1.0~10.0 mmol/L浓度区间内,随浓度增加和时间的延长,阿司匹林对细胞的抑制率逐渐增加。结论阿司匹林在体外可抑制人宫颈癌Hela细胞增殖,其机制可能为抑制肿瘤细胞DNA合成有关。
Objective To explore whether aspirin (acetylsalieylic acid, ASA ) could inhibit the growth of Hela cells of cervical cancer. Methods Hela cells were cultured in vivo and treated with ASA of different concentrations for 24 h to 96 h. The structures of cell were observed under light microscope with Giemsa stain. The cell proliferation inhibitory rates (CPIR) were tested by MTT method and the apoptosis ladder of DNA was evaluated by agarose gel electrophoresis. Results By Giemsa staining, Hela cells treated with aspirin were smaller and smaller, and apoptotic body also were observed. ASA inhibited Hela cell proliferation in a time and dose dependent manner. The apoptosis ladder of DNA was detected by DNA agarose gel electrophoresis alter treated with aspirin of various concentrations for 48 hours. Aspirin could inhibit the proliferation of Hela cell among concentrations of 1.0 - 10. 0 mmol/L As the dose and time increasing, the inhibitory of aspirin on proliferation of Hela cell was significantly increased. Conclusions ASA could inhibit the proliferation of Hela cells in vivo, and the possible mechanism is inhibiting the DNA synthesis.
