IL-17、RANTES及其受体在早期自然流产蜕膜组织中的表达及意义

摘要 目的检测IL-17、T细胞激活上调性表达分泌因子s(RANTES)及其受体CCR5在正常早孕及早期自然流产患者蜕膜组织中的表达,并探讨其意义。方法30例早期自然流产患者(流产组)和30例正常早孕妇女(对照组)的蜕膜组织,分别应用RT—PCR、ELISA方法检测蜕膜组织中IL-17、RANTES及CCR5的表达并进行线性相关分析,同时应用免疫组织化学方法染色定位其蛋白的表达。结果①早孕蜕膜组织中均有IL—17mRNA、RANTESmRNA、CCR5mRNA的表达,流产组蜕膜组织中IL-17mRNA、RANTESmRNA、CCR5mRNA的表达水平高于对照组,差异有统计学意义(P〈0.05)。②ELISA法定量检测结果与PT—PCR结果趋势一致。③IL-17与RANTES的表达呈正相关。④IL-17定位于蜕膜细胞胞质,RANTES定位于蜕膜细胞胞质及胞膜,CCR5定位于蜕膜细胞胞核。结论早期自然流产患者蜕膜组织中IL-17、RANTES及其受体CCR5的表达水平较正常早孕者高,这种改变可能是导致早期自然流产的重要原因。 Objective To investigate the expression of intedeukin - 17 (IL - 17 ), regulated on activation normal T cell expressed and secreted(RANTES) and CCR5 in decidua of first trimester spontaneous abortion and its significance. Methods Expression of IL - 17, RANTES and CCR5 in decidua of first trimester spontaneous abortion ( abortion group, n = 30) and normal first trimester pregnant women ( control group, n = 30 ) were detected by semi - quantitative RT - PCR, ELISA,and linear correlation analysis was performed. Its protein was detected by immunohistochemistry. Results (1)The IL - 17, RANTES and CCR5 mRNA gene expressions were detected in first trimester decidua tissues, expression of IL - 17, RANTES and CCR5 mRNA was significantly higher in abortion group compared with control group ( P 〈 0.05 ). (2)The ELISA quantitative test results were consistent with the RT - PCR results. (3)Expression of IL - 17 in de- cidua of first trimester was positively correlated with expression of RANTES. (4)IL - 17 existed in the decidual cell cyto- plasm, RANTES positioning on decidual cell nuclei, CCR5 located in the decidual cell cytosol and membrane. Conclu- sion The higher expression of IL - 17, RANTES and CCR5 in decidua of early spontaneous abortion may be the impor- tant cause leading to spontaneous abortion in early pregnancy.
