摘要
目的:探讨大鼠骨髓源性内皮祖细胞(EPCs)的培养与鉴定方法,以及电针血清对其增殖、迁移、黏附等功能的影响。方法:将SD大鼠随机分为正常组、模型组、电针组,后两组制造脑缺血再灌注模型后分别不治疗和采用电针治疗,48h后获取各组血清。将不同处理组的血清加至体外分离、培养、鉴定过的SD大鼠骨髓来源的EPCs,设对照组,分别采用MTT法、迁移实验、黏附实验观察电针血清对EPCs增殖、迁移、黏附能力的影响。结果:电针组EPCs在增殖能力方面OD值为(0.380±0.052),迁移细胞数为(46.00±1.32),黏附细胞数为(44.67±1.53)。与对照组相比,正常组、模型组、电针组增值、迁移、黏附能力均明显增强(P<0.05,P<0.01);与正常组相比,电针组与模型组增殖、迁移、黏附能力均显著增强(P<0.05,P<0.01);与模型组相比,电针组在增殖能力方面无明显差异,但迁移、黏附能力显著增强(P<0.01)。结论:电针血清能明显提高EPCs的增殖、迁移、黏附能力。
Objective: To study the culture and identification of sprague dawley (SD) rat bone marrow-derived EPCs, and investigate the effect of electroacupuncture serum on the proliferation, migration, adhesion of cultured EPCs. Methods: SD rats were randomly divided into normal group, model group, etectroacupuncture group. Except the normal group, the others were built middle cerebral artery occlusion model, and the electroacupuncture group were treated by electroacupuncture. After 48 hours, got every group serum at the same time, bone marrow mononuclear cells were isolated, cultured and identified, proved to be EPCs. Adding the different serum to the EPCs, using the MTT assay, migration assay, adhesion assay to observe the effect of electroacupuncture. Results: The electroacupuncture group's OD value was (0.380±0.052) in the proliferation, migration cells number was (46.00±1.32), adhesion cells number was (44.67±1.53). Compared with the control group, the normal group, model group and electroacupuncture group showed significant difference (P〈0.05, P〈0.01). Compared with the normal group, the model group and the electroacupuncture group were different significantly on the proliferation, migration, adhesion (P〈0.05, P〈0.01); Compared with the model group, electroacupuncture group was not significantly different in the proliferation, but the migration and adhesion were different significantly (P〈0.01). Conclusion: Electroacupuncture serum can obviously enhance the proliferation, migration, adhesion of EPCs.
