摘要
目的:研究金欣口服液含药血清对呼吸道合胞病毒(RSV)感染的小鼠肺泡巨噬细胞(RAW264.7)Toll样受体7(TLR7)信号转导通路的影响,探讨其可能的抗病毒机制。方法:RSV感染培养的RAW264.7细胞,用金欣口服液含药血清进行干预,24h后收集细胞及上清液,Western Blot法检测RAW264.7细胞TLR7、髓样分化因子-88(MyD88)、核因子(NF-κB)、干扰素调节因子7(IRF7)蛋白表达,ELISA法检测细胞上清液肿瘤坏死因子-α(TNF-α)、干扰素-α(IFN-α)的含量。结果:金欣口服液含药血清能降低RSV感染24h后TLR7、MyD88、NF-κB蛋白表达水平(P<0.01,P<0.05),而对IRF7的表达无明显的影响;下调细胞上清液中的TNF-α表达(P<0.01),IFN-α未能测出。结论:金欣口服液含药血清可能通过抑制TLR7/MyD88/NF-κB/TNF-α通路发挥抗RSV作用。
Objective: To study the effect of drug serum of Jinxin Oral Liquid on TLR7 signal transduction pathway in RAW264.7 infected with respiratory syncytial virus (RSV), and to explore its antiviral mechanism. Methods: RAW 264.7 cells were infected with RSV, and treated with the drug serum of Jinxin Oral Liquid. 24 hours later, cells were collected to detect expressions of protein TLR7, MyD88, NF-κB, IRF7 with Western blot assay, and the supernatant was collected to detect the level of TNF-a, IFN-tt with ELISA assay. Results: The drug serum of Jinxin Oral Liquid decreased the expression of TLR7, MyD88, NF-κB (P〈0.01, P〈0.05), but had no effect on the expression of IRF7. In the supernatant, the level of TNF-a was down regulated with the drug serum of Jinxin Oral Liquid (P〈0.01), but the level of IFN-a could not be detected. Conclusion: Anti-RSV effect of Jinxin Oral Liquid may be made through inhibiting TLR7/MyD88/NF-κB/TNF-a signal transduction pathway.
