摘要
目的:探讨不同减重术式对GK大鼠骨骼肌SIRT3酶表达的影响。方法:24只Goto-Kakizaki(GK)大鼠随机分为Roux-en-Y胃转流手术组(Roux-en-Y Bypass group RY组,n=8)、袖状胃切除手术组(Sleeve gastrectomy group SG组,n=8)、胃绑带手术组(Gastric banding group GB组,n=8),3组分别行相应术式,分别测定术前、术后48h和1、2及4周空腹血糖水平,术后4周处死大鼠取大腿部骨骼肌,用western blot法半定量测定其SIRT3酶值。结果:术前3组大鼠空腹血糖水平相仿,术后3组血糖均有不同程度下降,以RY组和SG组下降明显,RY组和SG组下降差异无统计学意义(P>0.05),与GB组相比差异有统计学意义(P<0.05),同样3组术后骨骼肌SIRT3酶表达均有增加,以RY组和SG组增加明显,RY组和SG组增加差异无统计学意义(P>0.05),与GB组相比差异有统计学意义(P<0.05)。结论:不同减重术式均可增加GK大鼠骨骼肌SIRT3酶表达,尤以Roux-en-Y胃转流手术和袖状胃切除手术对其影响明显。
Objective. To explore effect of different weight losing method on SIRT3 enzyme in skeletal muscle of Goto-Kakizaki (GK) rats. Methods. A total of 24 GK rats were randomly divided into Roux-en-Y (RY) group (n=8), sleeve gastrectomy (SG) group (n=8) and gastric banding (GB) group (n;8). Fasting blood-glucose level before surgery, 48 h, 1 week, 2 weeks, and 4 weeks after surgery were measured. Rats were killed 4 weeks after surgery, and skeletal muscle from huckle were taken. SIRT3 was detected by western blot. Results. There was no difference in fasting blood-glucose level among three groups before surgery. And the levels were decreased after surgery in three groups. The decrease was more significant in RY and SG group, but the difference was not significant between these two groups (P〈0.05). SIRT3 was increased after surgery in three groups. The increase was more significant in RY and SG group, but the difference was not significant between these two groups(P〈0.05). Conclusions. Three weight losing methods all can increase SIRT3 expression, and the effect of Roux-en-Y gastric bypass and sleeve gastrectomy are more significant.
