摘要
目的探讨甲状腺癌miRNA-146表达与BRAF基因突变之间的相关性。方法采用免疫组化法,对30例甲状腺癌组织、28例正常组织中miRNA-146表达水平进行检测,并根据染色程度和阳性细胞数进行综合评分;采用聚合酶链式反应(PCR)对甲状腺癌中BRAF突变进行检测,同时分析miRNA-146表达水平与BRAF突变之间的相关性。结果甲状腺癌组织30例中有27例(90.0%)出现miRNA-146表达阳性,而在正常组织中miRNA-146表达为阴性,2组染色评分均值分别为(0.07±0.03)分和(7.33±0.01)分,2组比较,差异具有统计学意义(t=4.303,P<0.05);在甲状腺癌患者中,BRAF突变阳性组miRNA-146表达明显高于阴性组,miRNA-146表达评分均值分别为(9.51±0.29)分和(4.45±0.39)分,2组比较,具有统计学差异(t=2.920,P<0.05)。结论 miRNA-146在甲状腺癌中表达增加,且其表达水平与BRAF突变密切相关。
Objective To discuss the correlation between thyroid cancer miRNA-146 expression and BRAF gene muta- tion. Methods Immunohistochemistry method was used to detect the expression level of miRNA-146 in 28 cases of normal tis- sues and 30 cases of thyroid carcinoma tissues, and comprehensive score was evaluated according to the number of positive cells and staining degree. Polymerase chain reaction(PCR) below the mutations were detected by BRAF in thyroid cancer, and correlation between miRNA-146 expression levels and BRAF mutation was analyzed. Results Out of the 30 cases of thyroid carcinoma,27 cases(90.0% ) showed miRNA-146 positive expression, while miRNA-146 expression in normal tissues was negative. Staining scores in the two groups were 0.07 ± 0.03 and 7.33 ± 0.01 respectively. The difference was statistically sig- nificant ( t = 4. 303,P 〈 O. 05 ). In patients with thyroid cancer, the expression of miRNA-146 in BRAF mutation positive group was significantly higher than that in the negative group. The mean scores for miRNA-146 expression were 9.51 ± 0.29 and 4.45± 0.39 respectively. The comparison between the two groups showed statistical difference ( t = 2. 920 ,P 〈 0.05 ). Conclu- sion Increased expression of miRNA-146 in thyroid carcinoma, and the expression level of BRAF are closely related with mutation.
