Disregulation in Th1/Th2 Subsets of CD4+ T Cells in Peripheral Blood of Soft Tissue Sarcoma Patients

摘要 目的 :观察软组织肉瘤患者T淋巴细胞亚群中Th1/Th2的变化 ,为细胞因子免疫治疗提供依据。方法 :应用放射免疫分析法以及酶联免疫分析法检测 10例软组织肉瘤患者及 10例健康志愿者血清中IL 2、IL 4、IL 6、IL 10、IL 12的含量 ,检测外周血单个核细胞培养上清中IL 2、IL 4含量。结果 :软组织肉瘤患者血清及培养上清中IL 2、IL 12减少 ,而IL 10、IL 4、IL 6含量增加。经过植物血凝素刺激的培养淋巴细胞上清液中的IL 4含量显著增加 ,而IL 2显著降低。结论 :软组织肉瘤患者体内存在Th1/Th2平衡失调 ,其中Th1亚群功能抑制 。 objective: To study the change of TH1/TH2 subjects of CD4+T cell in peripheral blood of soft tissue sarcoma patients. Methods:Serum levels of IL 2,IL 4, IL 6, IL 10, IL 12 were examined in 10 patients and 10 healthy subjects by RIA and ELISA. The production of IL 2, IL 4 , with or without PHA by PBMC , also were measured . Result: In the serum and medium supernatant fluid from the patients with soft tissue sarcoma,the levels of IL 2,IL 12 decreased,but the levels of IL 4,IL 6,IL 10 increased,while after stimulation of mitogen PHA,the content of IL 4 in the supernate of cultured lymphocyte was significantly increased,but the level of IL 2 prominently decreased.Conclusion:In the peripheral blood of patients,there seems to be a disregulation to the functions of Th1/Th2 subsets of CD4+ T cell with a hyperfunction of Th2 and inhibition of Th1 cell,it suggests the existence of soft tissue sarcoma in the host
