摘要
以灰毡毛忍冬嫩茎为外植体进行组织培养,优化并建立了最佳外植体组培体系,对灰毡毛忍冬不同组培阶段的最适培养基进行了筛选。结果表明:外植体用70%乙醇10 s+10%过氧化氢溶液10 min消毒处理,污染率最低;最适初代培养基为N6+1.0mg/L BA+0.1 mg/L NAA,外植体的分化率可达93%;最佳继代增殖培养基为N6+1.5 mg/L BA+0.5 mg/L NAA,灰毡毛忍冬新生组培苗生长旺盛,增殖系数高达5.2;最佳生根培养基为1/2MS+1.5 mg/L IBA+1%活性炭最好,试管苗生根率及驯化移栽成活率均达95%以上。
Tender stems of Lonicera macranthoides Hand. Mazz were taken as explants to conduct tissue culture. The optimal explant tissue culture system was established, and the optimum culture media for different periods of tissue culture of Lonicera macranthoides Hand. Mazz were selected. The results showed that the contamination rate was minimum when treated explants with 70% C2HsOH for lOs and with 10% H202 for 10 min; the optimum initial culture medium was N6+l.0 mg/L BA+O.10 mg/L NAA, in which the regeneration ratio of explants reached 93%; the optimal subculture medium was N6+1.50 mg/L BA + 0.5mg/L NAA, in which the regenerated seedhngs of Lonicera macranthoides Hand. Mazz grew strong and its proliferation rate reached 5.2 times; the optimal rooting medium was 1/2 MS+I.50 mg/L IBA with 1% activated carbon, in which the rooting rate and the survival rate of regenerated seedlings were more than 95%.
