摘要 目的研究槲皮素(QUE)对肺腺癌A549/DDP细胞多药耐药(MDR)的逆转作用及其对P-糖蛋白(P-gp)及survivin蛋白表达的影响。方法四甲基偶氮唑蓝(MTT)法研究QUE对肺腺癌A549/DDP细胞MDR逆转作用;流式细胞法(FCM)分析QUE对P-gp、survivin蛋白表达的影响。结果浓度为10μg.ml-1的QUE使A549/DDP细胞对顺铂(DDP)的半数抑制浓度(IC50)由(31.34±2.99)μg.ml-1降至(9.09±1.23)μg.ml-1,P<0.01,逆转倍数(RF)为3.45。A549/DDP细胞均高表达P-gp及survivin蛋白,平均荧光指数(AFI值)分别为4.42和3.96,两蛋白的表达水平均随QUE浓度增加而呈逐渐下降趋势。结论 QUE对A549/DDP细胞MDR有明显的逆转作用,其作用与P-gp及survivin蛋白表达下降有关。 Objective To evaluate the effect of quercetin ( QUE ) on reversal multidrug resistance (MDR) of A549/DDP cells, and explore the expression of P-gp and survivin interfered by QUE. Methods Reversal effect of different concentrations of QUE on MDR of A549/DDP cells were tested by MrIT assay. Flow cytometry ( FCM ) was used to investigate the impacts on P-gp and survivin expres- sions of A549/DDP cells. Results QUE had obvious effect on MDR reversal of A549/DDP ceils. The ICS0 was (9. 09 ± 1.23 )μg · m1-1 ,which had obviously difference to control group (31.34 ±2.99) μg · m1-1 (P 〈0. O1 ). After interfering by QUE with different concentrations, the expression of P-gp and survivin in cells became fewer, which had significant difference to control group ( P 〈 0.05, P 〈 0. 01 ). Conclusion QUE could reverse MDR of A549/DDP cells, which might have relation with down-regulating expressions of P-gp and survivin.
