丹参总酚酸类成分的提取纯化工艺和质量研究 被引量:8

摘要 目的:提取纯化丹参中水溶性酚酸类有效成分,并对其进行质量分析。方法:以HPLC检测丹酚酸B的含量,采用单因素和正交试验优化丹参的提取工艺;采用大孔吸附树脂法纯化丹参水溶性成分,并对丹参及其提取物的水溶性成分进行HPLC指纹图谱研究,制定相应质量标准。结果:丹参总酚酸提取物中主要水溶性成分丹酚酸B的提取转移率达85%,提取纯化物中含丹酚酸B>65%,其他丹酚酸12%;相应HPLC指纹图谱分析方法可有效分析和鉴别主要水溶性成分的特征峰。结论:该方法能够快速高效分离纯化丹参水溶性成分,得到较高纯度的丹参总酚酸;HPLC指纹图谱分析方法的精密度、重复性良好。 This study was aimed to extract and purify active components of Salvia miltiorrhiza and to determinate the relevant quality analysis. The high-performance liquid chromatography (HPLC) method of salvianolic acid B was established. Factors influencing the extraction were investigated with a single-factor and orthogonal design. The water-soluble components of S. miltiorrhiza were purified with macroporous resin. The HPLC fingerprint of S. miltiorrhiza and its purified extract were studied, and the quality specification was established. The results showed that the transfer rate of salvianolic acid B was up to 85%, contents of salvianolic acid B were 65%, and other salvianolic acid is about 12%. The HPLC fingerprint analysis method can effectively analyze and identify the main characteristic peaks of water-soluble components. It was concluded that the method can quickly and efficiently purify S. miltiorrhiza water-soluble components. The high-purity salvianolic acid components were obtained. The HPLC fingerprint method has good precision and reproducibility.
