氟西汀对不同品系小鼠悬尾实验的影响 被引量:2

Strain-dependent effects of fluoxetine on tail suspension test in mice

摘要 目的研究抗抑郁药氟西汀对小鼠悬尾实验的影响是否有品系差异性。方法选择2个远交系(昆明和ICR)小鼠,3个近交系(C57BIZ6,Balb/c,DBAZ2)小鼠进行悬尾实验,给予选择性5-羟色胺再摄取抑制剂(SSRI)氟西汀,观察对悬尾不动时间的影响。结果小鼠悬尾不动时间基础值有明显差异,其中时问最长的是C57BL/6小鼠[(145.0±16.8)S],而最短的是DBA/2小鼠[(34.5±6.1)S]。氟西汀能显著降低C57BIM6[给药组(116.0±10.3)S,对照组(145.0±16.8)S]、Balb/c[给药组(44.3±6.2)S,对照组(75.3±10.3)s]和DBA/2小鼠[给药组(16.6±4.3)S,对照组(34.5±6.1)S]的悬尾不动时问(p〈0.05);而昆明和ICR小鼠的不动时间在给予氟西汀后没有显著变化。结论小鼠悬尾实验有明显的品系差异,氟西汀在C57BIM6、Balb/c和DBA/2小鼠上显示抗抑郁作用,而对昆明和ICR小鼠无明显抗抑郁效应。 Objective To study the strain difference in response to the antidepressant fluoxetine in mouse tail suspension test. Methods Two outbred mouse strains (KM and ICR) and three inbred mouse strains (C57BIJ6, Balb/e and DBA/2 ) were used in this study. They were treated with the selective serotonin reuptake inhibitor (SSRI) fluoxetine or saline and the immobility time in tail suspension test was recorded. Results There was significant difference of baseline inunobility time among different stains with C57BL/6 the most immobile( ( 145.0 ± 16.8 )s ) and DBA/2 the least (( 34.5 ± 6. 1 )s ). Fluoxetine significantly decreased the immobility time in C57BL/6 ( ( 116.0 ±10.3 ) s vs ( 145.0 ± 16.8 ) s) of control), Balb/c ( (44.3 ± 6.2) s vs ( 75.3 ± 10.3 ) s ) of control ) and DBA/2 mice ( ( 16.6 ± 4.3 ) s vs ( 34.5 ± 6. 1 ) s ) of control ),while the immobile time of KM and ICR mice was not influenced by fluoxetine. Conclusion The effects of fluoxetine in tail suspension test are strain dependent. Fluoxetine exhibits anti- depressant effects in C57BL/6,Balb/c and DBA/2 mice,but not in KM and ICR mice.
