胃癌区域转移淋巴结中锌指蛋白139与基质金属蛋白酶-2、基质金属蛋白酶-7、基质金属蛋白酶组织抑制剂-1表达的关系及意义 被引量:18

Relationship between zinc finger protein 139 with matrix metalloproteinase-2, matrix metalloproteinase-7 and tissue inhibitor of metalloproteinase-1 in local lymph node metastases of gastric cancer

摘要 目的检测锌指蛋白139(ZNF139)与基质金属蛋白酶(MMP)-2、MMP-7、基质金属蛋白酶组织抑制剂(TIMP)-1在胃癌原发灶、区域淋巴结转移灶中的表达,并探讨其意义。方法应用逆转录-聚合酶链反应(RT—PCR)及免疫组织化学链菌素抗生物素蛋白-过氧化物酶(SP)法检测46例进展期胃癌患者的原发灶和淋巴结转移灶中ZNF139、MMP-2、MMP-7、TIMP-1mRNA和蛋白水平。结果胃癌区域淋巴结转移灶中ZNF139、MMP-2、MMP-7mRNA表达分别为0.63±0.09、0.75±0.11、0.73±0.11,蛋白阳性率分别为80.43%、84.78%、86.96%,均明显高于原发灶的mRNA(0.32±0.05、0.27±0.04、0.21±0.03)和蛋白阳性率(54.35%、52.17%、58.70%);而转移灶TIMP-1mRNA(0.63±0.09)和蛋白阳性率(43.48%)则明显低于原发灶(1.24±0.18、78.26%,P〈0.05)。ZNF139与MMP-2、ZNF139与MMP-7的在原发灶中mRNA表达均呈正相关(r分别为0.3258、0.3446,P〈0.05),ZNF139与TIMP-1无明显相关(P〉0.05);ZNF139与MMP-7在淋巴结中mRNA表达水平呈正相关(r=0.3866.P〈0.05),而ZNF139与MMP-2、ZNF139与TIMP-1无明显相关(P〉0.05)。结论ZNF139可能通过调节部分侵袭相关基因的表达参与了胃癌的侵袭转移。 Objective To investigate the expressions and significance of zinc finger protein 139 (ZNF139) , matrix metalloproteinase (MMP) -2, MMP-7, tissue inhibitor of metalloproteinase (TIMP) -1 in primary tumors (PT) and local lymph node metastases (LNMs) of gastric carcinoma. Methods The expression levels of ZNF139, MMP-2, MMP-7 and TIMP-1 were detected by using reverse transeriptasepolymerase chain reaction (RT-PCR) and immunohistochemistry in 46 paired PT and LNMs of gastric cancer. Results The expression levels of ZNF139, MMP-2 and MMP-7 mRNA (0. 63 ± 0. 09, 0. 75 ± 0. 1 1, 0. 73 ± 0. 11 ) and positive rate of proteins (80. 43% , 84. 78% , 86. 96% ) were all higher in LNMs than the mRNA expression levels (0. 32 ±0. 05, 0. 27 ±0. 04, 0. 21 ±0. 03) and positive rate of proteins (4. 35% , 52. 17% , 58.70% ) in PT, whereas the expression of TIMP-1 mRNA and positive rate of protein were lower in LNMs (0. 63 ± 0. 09, 43.48% ) than in PTs ( 1.24 ± 0. 18,78.26% ) ( all P 〈 0. 05 ). In PT there was a positive correlation between ZNF139 and MMP-2 or MMP-7 mRNA (r = 0. 3258, 0. 3446, both P 〈 0. 05 ). In LNMs, positive correlation was found between ZNF139 and MMP-7 (r = 0. 3866,P 〈 0. 05). Conclusion ZNF139 may be invovled in invasion and metastasis of gastric cancer by regulting some genes.
