钩藤和莱菔子生物碱抗高血压血管内皮细胞损伤效应 被引量:36

Protective effect of total alkaloids from Uncariae Ramulus cum Uncis combined with Raphani Semen on vascular endothelial cell injury by hypertension

摘要 目的探讨钩藤总生物碱配伍莱菔子水溶性生物碱抗高血压大鼠血管内皮细胞损伤的效应及初步机制研究。方法采用腹腔注射N-硝基左旋精氨酸(L-NNA)方法制备大鼠高血压模型;采用无创性套尾法测定大鼠清醒状态下尾动脉收缩压;采用酶联免疫吸附法测定大鼠血浆血管性血友病因子(vWF)、内皮素-1(ET-1)、细胞间黏附分子1(ICAM-1)、血管细胞黏附分子1(VCAM-1)和P选择素(P-S)。结果钩藤总生物碱配伍莱菔子水溶性生物碱对L-NNA诱导的高血压大鼠有显著降压效用(P<0.05),并能降低高血压大鼠血浆vWF、ET-1、ICAM-1、VCAM-1和P-S水平(P<0.05)。结论钩藤总生物碱配伍莱菔子水溶性生物碱具有良好降压和血管内皮保护效应,其机制与抑制血管内皮细胞分泌黏附因子、减轻血管壁炎症反应有关。 AIM To study the protective effect and initial mechanism of total alkaloids from Uncariae Ramulus cum Uncis combined with total alkaloids from Raphani Semen on vascular endothelial cell injury by hypertension. METHODS The rat model for hypertension w~/s established by injection of N'-nitro-L-arginine (L-NNA) in ab- dominal cavity. Systolic blood pressure was measured by non-invasively tail-cuff technique when the rats were in a- wakened state. Plasma yon Willebrand factor (vWF), endothelin-1 (ET-1), intercellular adhesion molecule-1 (ICAM-1), vascular cellular adhesion molecule-1 (VCAM-1) and P-selection (P-S) were measured by enzyme- linked immunosorbent assay. RESULTS Total alkaloids from Uncariae Ramuluz cum Uncis combined with total alkaloids from Raphani Semen significantly declined the blood pressure of hypertension rats induced by L-NNA (P 〈0.05). At the same time, the plasma levels of vWF, ET-1, ICAM-1, VCAM-1, P-S decreased significantly (P 〈 0.05 ). CONCLUSION Total alkaloids from Uncariae Ramulus cure Uncis combined with total alkaloids from Raphani Semen have effective antihypertensive function related to restrain the adhesion molecules which secrete by vascular endothelial cell and alleviate inflammatory reaction of vascular wall.
