摘要
采用腹腔注射交感神经化学切断剂6-羟多巴胺(6-OHDA)的方法,在小鼠束缚应激模型上观察外周交感神经在应激免疫抑制蛋向产生中的作用。结果表明,腹腔注射6-OHDA后,血清中应激免疫抑制蛋白的产生明显减少,第五天降至最低点,以后逐渐恢复,第 14天恢复正常。脾脏和淋巴结中去甲肾上腺素( NE)的含量,在注射6-OHDA后第一天明显降低,而且至少在14天内维持在很低水平。说明6-OHDA已损坏了交感神经纤维。注射6-OHDA前30min注射去郁敏(DMI),阻断交感神经的重吸收作用,可明显对抗6-OHDA抑制应激免疫抑制蛋白产生作用。以上结果表明,外周交感神经具有促进应激免疫抑制蛋白生成的作用。
In the present study 6-hydroxydopamine(6-OHDA) was used for chemical sympathectomy and the effect of 6-OHDA on generation of stress-induced immune suppressive protein induced by restraillt stress was studied. It was found that the generation of lSPS was significantly suppressed. The maximum suppression was found on day 5 after 6-OHDA injection and it recovered gradually to normal on day 14. In order to assess the effectiveness of the 6-OHDA injection, the contents of norepinephrine(NE) in spleen and lymph node were assayed by ligh performance liquid chromatography with electrochemical detector (HPLC-ED). NE was found significantly deereased on dey 1 and the content was at the very low level at least for 14 days. The toxic effect of 6-OHDA on sympathetle nervous fibers was known the selective uptake of 6-OHDA into sympathetic nervous fibers. Desipramine(DMI) was reported to be able to block 6-OHDA uptake.In control esperiment in our study, rats were injected with DMI 30 min before the injection of 6-OHDA. The resutts showed that the antagonist effect of 6-OHDA on the generation of ISPS was blecked by the injection of DMI.Taken together, the results obtained in the present study illustrate that on the condition of restraint stress peripheral sympathetic nervous fiber plays an important role in mediating the generation of IPSP.
