摘要
目的:建立HPLC同时分离测定了核桃仁中胞苷、尿苷、鸟苷、腺苷的含量。方法:采用Agilent TC-C18色谱柱(4.6 mm×250 mm,5μm),以甲醇-10 mmoL.L-1乙酸铵为流动相梯度洗脱,流速0.5 mL.min-1,检测波长260 nm,柱温30℃,测定了10个产地核桃仁中胞苷、尿苷、鸟苷、腺苷的含量。结果:4种核苷类成分在各自线性范围内,峰面积值与进样量呈良好的线性关系(r=0.999 3~0.999 9);回收率为98%~101%(RSD<3%),结论:方法简便快速、结果准确、重复性和稳定性好,可为核桃的含量测定提供参考。
Objective: To determine the content of cytidine, uridine, guanosine and adenosine in walnut kernel by HPLC. Method: The HPLC method was used to determine respectively the content of cytidine, uridine, guanosine and adenosine in walnut kernel from 10 different regions. The analysis was carried out on Agilent TC-C18 column (4.6 mm×250 mm, 5 μm). Methanol-10 mmoLL-1ammonium acetate was used as mobile phase with gradient elution; the flow rate was 0.5 mLmin-1, detection wavelength was set at 260 nm and the column temperature was kept at 30 ℃. Result: The injection amounts of four nucleosides compounds over respective ranges displayed good linear relationship with the peak area and the correlation coefficients equaled or exceeded 0.999 3.The average spike recoveries for four nucleosides compounds ranged from 97% to 102%, with a RSD of less than 3%. Conclusion: The method is simple, quick, accurate and reproducible, and can be provide reference for the quality control of the walnut kernel.
