摘要
[目的]分析印记基因SNRPN、NDN和UBE3A在初生仔猪中的表达情况,从而探讨它们和猪生长发育的关系。[方法]采用荧光定量PCR方法,检测SNRPN、NDN、UBE3A基因在出生3 d仔猪的13种组织(心、肝、脾、肺、肾、肌肉、大脑、垂体、小肠、胃、膀胱、脂肪、下丘脑)及不同时期的胎盘中的表达水平。[结果]SNRPN、NDN、UBE3A基因在13种组织中都有表达,SNRPN和NDN基因在下丘脑中表达量极显著(P<0.01)高于其他被测组织,UBE3A基因在肺、小肠和下丘脑中的表达量显著(P<0.05)高于其他被测组织,SNRPN基因在下丘脑中的表达量极显著(P<0.01)高于NDN、UBE3A基因,在梅山猪和杜洛克猪不同时期胎盘中稳定表达。[结论]SNRPN、NDN和UBE3A基因都在下丘脑中高表达,对神经系统的生长发育有重要的影响;同时该研究为这3种基因在猪中的功能研究提供了基础。
[Objective] The object was to analyze the expression profile of SNRPN,NDN and UBE3A in neonatal piglets and explore the relationship with growth and development.[Method] mRNA levels of the three imprinted genes were detected using quantitative real-time PCR(qRT-PCR).[Result] The imprinted genes of SNRPN,NDN and UBE3A were expressed in all the tissues.Among them,SNRPN and NDN had higher mRNA level in hypothalamus than in other tissues(P<0.01).UBE3A also had higher mRNA level in hypothalamus,in addition to the lung and small intestine(P<0.05).Moreover,SNRPN had highest mRNA level than NDN and SNRPN in hypothalamus(P<0.01),and was stable expressed in Meishan and Duroc different period of placentas.[Conclusion] SNRPN,NDN and UBE3A all had the high level of expression in hypothalamus.The results showed that the three genes were related to the growth and development of the nervous system.The result of this study will lay the foundation for the further research of functions and mechanisms in pig in the future.
