五子衍宗复方改善Sertoli细胞氧化应激损伤的作用

Eeffect of Wuziyanzong compound recipe on oxidative stress injury and apoptosis of sertoli cells

目的探讨五子衍宗复方对Sertoli细胞氧化应激损伤的改善作用。方法将TM4细胞培养液分为空白组、模型组、低、中、高剂量加药组分别给予相应药物处理。酶标仪上检测各组的吸光度值,硫代巴比妥酸比色法测定SOD活性和黄嘌呤氧化酶法测定MDA含量,流式细胞仪测定各组的细胞凋亡率和存活率,PCR检测caspase-3 mRNA的表达。结果五子衍宗复方可提高TM4细胞的活性,增加SOD活性,降低MDA含量,能降低细胞的凋亡率,并提高细胞的存活率,抑制细胞内caspase-3 mRNA的表达,且呈剂量依赖性。结论五子衍宗复方能改善Sertoli细胞的氧化应激损伤,提高细胞的存活率。

Objective It is to approach the effect of Wuziyanzong compound recipe on oxidative stress injury and apoptosis of sertoli cells. Methods TM4 cells were divided in control group, model group, and low, medium and high dose group, and were treated with homologous drug. The absorbance values of each group were detected on ELIASA ; the SOD activity was detected with Penthiobarbital acid colorimetry technique, the MDA content was detected xanthine oxidase technique, the apopto- sis and survival rates of cells were detected by flow cytometry, the caspase - 3 mRNA expression were detected by real time - PCR. Results Wuziyanzong compound recipe had significantly enhanced the activity of TM4 cells, increased SOD activity and decreased MDA content, reduced the rate of apoptosis and enhanced cell survival, inhibited the expression of caspase -3 mRNA in the cells. The effect was dose-dependent. Conclusion Wuziyanzong compound recipe can improve the oxidative stress injury and enhance cell survival for sertoli cell.
