摘要
目的 研究海马NMDA受体 2A亚基 (NR2A)在脑缺血 再灌注过程中酪氨酸磷酸化水平的变化。方法采用SD大鼠四动脉结扎建立前脑缺血模型。用抗磷酸酪氨酸单抗或抗NR2A抗体以免疫沉淀法和免疫印迹法检测海马NR2A酪氨酸磷酸化水平。结果 缺血海马NR2A酪氨酸磷酸化水平下降 ,再灌注后升高 ,再灌注 6h达最高 ,其后又下降。缺血再灌注后NR2A含量逐渐下降。结论 脑缺血
Objective To investigate the tyrosine phosphorylation of NMDA receptor subunit 2A (NR2A) after cerebral ischemia and reperfusion. Methods Four-vessel occlusion ischemic model of SD rats was used in this study. Tyrosine phosphorylation of NR2A in hippocampus was assessed by immunoprecipitation and immunoblotting using anti-phospho-tyrosine antibody and antibody to NR2A. Results The tyrosine phosphorylation of NR2A decreased during ischemia, increased after reperfusion, and reached its peak level at about 6 h after reperfusion. Then the phosphorylation decreased again. After reperfusion, the hippocampal level of NR2A decreased gradually. Conclusion Cerebral ischemia and reperfusion affect the NMDA receptor function through the changes of the tyrosine phosphorylation of NR2A.
