摘要
目的氧化锌代替铅丹制备无铅膏药基质,分步提取丁桂膏处方中的有效成分,最后混合滩涂制成无铅丁桂膏。方法按L9(3^4)正交试验表优选膏药基质的无铅化工艺,以水蒸气蒸馏法提取丁香、肉桂、白芷中的挥发油,以60—80%的乙醇提取川乌、草乌药材及丁香、肉桂、白芷药渣中的醇溶性成分并浓缩成稠膏,乳香、没药、樟脑、冰片直接粉碎成细粉最后加入。结果正交试验显示无铅膏药基质最佳工艺条件为麻油于330℃下熬炼3h,氧化锌与麻油高温聚混物配比(g/mL)为1:5,反应时间为20min。挥发油的最佳提取工艺为加10倍量水提取4.5h。醇溶性成分的最佳提取工艺为加入10倍量的70%的乙醇回流提取60min。结论药物有效成分与基质混合后制成的无铅丁桂膏,软化点和持黏力适宜,能代替黑膏药发挥治疗作用。
Objective To preparate lead-free plaster matrix of zinc oxide instead of read lead,to extracte the active ingredients in Dinggui prescription step by step,mix these active ingredients and cast into plaster. Methods Optimizating lead-free process by orthogonal table L9(34),extracting the volatile oil in the Cloves, Cinnamon,Angelica by steam distillation,and collecting alcohol-soluble components in Monkshood,Aconitum,Cloves, Cinnamon,Angelica by 60% to 80% ethanol,then condensed into thick paste,the Frankincense, Myrrh, Camphor, Borneol directly crushed into fine powder to join into the paste lastly. Results The optimum conditions of the lead-free plaster matrix showed in the orthogonal test were as follows:sesame oil was refined at 330~C for 3h,the ratio of zinc oxide(g)and high-temperature reaction mixture of sesame oil(ml) was 1:5,and the reaction time was 20min. The optimumum extraction of the volatile oil was that the amount of water was 10 times the herbs weight and the extraction time was 4.5h. The optimumum extraction of the alcohol-soluble components was that the ethanol concentration is 70% the volume of ethanol was 10 times the herbs weight and the extraction time was 60min. Conclusion The lead-free Dinggui plaster had suitable softening point and adhesive power can play a therapeutic role insiead of black plaster.
