Determination of Forchlorfenuron in Myica rubra by Matrix Solid Phase Dispersion Extraction and Reversed High Performance Liquid Chromatography

摘要 建立了基质分散萃取——反相高效液相色谱法测定杨梅中氯吡脲的方法:以杨梅为研究材料,乙腈为提取剂,甲醇为淋洗液、经氨基柱净化后,高效液相色谱检测,结果表明,以乙腈-水为流动相梯度洗脱,流速0.5 mL/min,经Kromasil C18(250 mm×4.6 mm,5μm锈钢柱)、UV 265 nm进行待测组分的分离和测定效果最佳,检测线性范围0.025~100μg/mL,相关系数r=0.999 9,添加低、中、高浓度的氯吡脲回收率分别为110%、108%、98.4%,相对标准偏差分别为2.6%、1.1%、0.97%,方法检出限为0.01 mg/kg。该方法测定杨梅中氯吡脲含量准确可靠。 Forchlorfenuron in Myica rubra was determined by matrix solid phase dispersion extraction (MSPD) and reversed high performance liquid chromatography(RP-HPLC). Myica rubra as the research material was extracted with acetonitrile, amino column was chosen for purifying column eluted by methanol and detected by HPLC. The results showed that HPLC operated under optimized flow phrase (acetonitrile-water) gradient elution and stainless steal Kromasil C18 column(250 mm ×4.6 mm,5 μm) ,and the flow rate was 5. 0 mL/min, the UV detection wavelength was 265 nm, quantified by external standard calibration. The linearity was good when the concentration ranged from 0. 125 to 100 μg/mL( r =0.999 9) . The recoveries of forchlorfenuron of low,medium and high concentration were 110%, 108% ,98.4%. And the relative standard deviations were 2.6% ,1.1% ,and 0.97% respectively. The limit of detection was 0.01 mg/kg. The method is accurate and reliable for determination of forchlorfenuron content in Myica rubra.
