赤雹根总皂苷对AA大鼠血清TNF-α与IL-10含量的影响

EFFECTS OFTOTALSAPONINS OFTHLADIANTHA DUBIA ROOT ON TNF-ΑAND IL-10 CONTENT IN SERUM OFADJUVANTARTHRITIS RATS

目的:通过观察赤雹根总皂苷(TSTDR)对佐剂性关节炎大鼠血清炎性细胞因子TNF-α与IL-10含量的影响,探讨TSTDR治疗类风湿性关节炎的作用机制。方法:60只大鼠随机分组:正常对照组,模型组,TSTDR低、中、高剂量组和雷公藤多苷组,每组10只大鼠。建立佐剂性关节炎大鼠模型后,TSTDR低、中、高剂量组大鼠分别灌胃给予不同剂量的TSTDR(40mg/kg/d、80mg/kg/d和160mg/kg/d),连续21d;雷公藤多苷组大鼠灌胃给予雷公藤多苷(12mg/kg/d),时间同TSTDR组大鼠。分别计算各组大鼠的关节炎指数,ELISA法检测各组大鼠血清TNF-α和IL-10的含量。结果:TSTDR各剂量组大鼠的关节炎指数、血清TNF-α含量显著低于模型组(P<0.01,P<0.05),TSTDR各剂量组大鼠血清IL-10含量显著高于模型组(P<0.01)。结论:TSTDR对大鼠佐剂性关节炎具有治疗作用,机制可能与降低促炎性因子TNF-α水平、提高炎性抑制因子IL-10水平有关。

Objective：To investigate the mechanisms of total saponin of Thladiantha dubia root （TSTDR） for treating rheumatoid arthritis by observing effects of TSTDR on TNF-α and IL- 10 content in serum of adjuvant arthritis rats.Methods： 60 rats were randomly divided into 6 groups with 10 rats in each group： normal control group, model group, TSTDR low dose group, TSTDR middle dose group, TSTDR high dose group and lripterygium glycosides group. After establishing adjuvant arthritis rats model, the tats in 3 TSTDR groups were lavaged TSTDR in different dose for 21 days （40mg/kg/d, 80mg/kg/ d, 160mg/kg/d）; the mrs in lripterygium glycosides group were lavaged Iripterygium glycosides （12mg/kg/d） for 21 days. The arthritic index of rats in each group was calculated; the TNF-α and IL-10 content in senma of rats in each group were detected respectively by ELISA.Results：The arthritic index and TNF-α content in serum of mrs in 3 TSTDR groups were obviously lower than that of rats in model group （P〈 0.01, P〈 0.05）; The IL-10 content in serum of rats in 3 TSTDR groups were obviously higher than that of rats in model group （P 〈 0.01 ）.Conclusions： TSTDR has therapeutic effects on adjuvant arthritis, the mechanisms maybe relate to decrease proinflammatory cytokines-TNF-α and increase anti-inflammatory cytokines-IL- 10.
