摘要 目的:了解慢性粒细胞白血病(CML)患者杀伤细胞免疫球蛋白样受体(killer cell immunoglobulin-like receptor,KIR)基因型及单倍型,为探讨KIR在慢性粒细胞白血病中发挥的作用奠定基础。方法:采用PCR-SSP方法对84例慢性粒细胞白血病患者进行KIR基因分型。结果:框架基因2DL4、3DL2、3DL3及3DP1存在于所有个体中,其他较为高频的基因依次为2DL1、2DL3、2DP1及3DL1,表型频率均>80%。CML患者的KIR2DL2和KIR2DS2表型频率较健康人有所降低,但除2DL5B(OR=0.208,P<0.05)外,分析的所有基因在CML患者和健康人间的分布均不具有显著差异。结论:基因KIR2DL5B可能与CML之间存在相关性。 Objective:To investigate the killer immunoglobulin-like receptor (KIR) genotype in patients with chronic myelogenous leukemia (CML). Methods:The sequence specific primer polymerase chain reaction (SSP-PCR) was employed to identify the KIR genes and pseudogenes in 84 CML patients. Results:The most frequent genotypes were KIR frame genes,including 2DL4,3DL2,3DL3 and 3DP1 (100%),followed by 2DL1,2DL3,2DP1 and 3DL1 (all above 80%). The frequencies of 2DL2 and 2DS2 in CML patients were lower than those in the control group,but there were no statistically difference between the two groups except 2DL5B (OR = 0.208,P 〈 0.05). Conclusion: KIR 2DL5B may be associatied with CML.
