锦鲤疱疹病毒-CJ株ORF124基因的克隆及生物信息学分析 被引量:3

摘要 为研究我国锦鲤疱疹病毒的基因背景信息,从锦鲤疱疹病毒中国吉林株(KHV-CJ株)获得ORF124基因,应用生物信息学方法分析KHV-CJ株ORF124基因结构和功能,并与Genbank上已公布的三株KHV进行比较分析。结果显示,ORF124基因的理论分子质量为31221.96 Da,KHV-CJ株与其他三株KHV同源性均为99%;信号肽切割部位最可能位于24位的T(苏氨酸)和25位的V(缬氨酸)氨基酸之间;ORF124基因无跨膜区;氨基酸序列不存在潜在的N-糖基化位点、11个潜在的O-糖基化位点和7个磷酸化位点。该蛋白的表面抗原决定簇位点区域广泛、峰值高,预示该基因可作为基因工程疫苗的重要候选基因。 To research genetic background information of koi herpesvirus China, the ORF124 gene was obtained from koi herpesvirus china Jilin strain which was isolated and saved in the laboratory. The structure and function of the gene was studied out by means of bioinformatics analysis and comparised analysis with three KHV strain gene had been published in genbank. The results showed that the molecular weight of ORF124 gene of koi herpesvirus China Jilin strain were 31221.96 Da, with the three KHV strain homology was 99 %. The most likely signal peptide cleavage site was between 24 bit (Thr) and 25 bit(Val). ORF124 had not transmembrane regions; amino acid sequence had not N - glycosylation sites, but had 11 potential O - glycosylation sites and 7 potential phosphorylation sites, the protein surface antigenic determinant was located in the extensive area , peak was high, and better than that of Jilin strains ( KHV - CJ strain) ORF81 and ORF83, it indicate that ORF124 gene could be used as an important candidate genes of genetic engineering vaccine.
