DMSO诱导的HL-60细胞分化后胞液Ca^(2+)浓度、磷酸化酶a和微粒体Ca^(2+)-ATP酶活性的改变

摘要 Ca^(2+)与肿瘤代谢有着极为密切的关系。本文测定了HL—60细胞经DMSO诱导分化前后胞液Ca^(2+)浓度、磷酸化酶a活性和微粒体Ca^(2+)—ATP酶活性。结果表明,DMSO加入HL—60细胞培养液后120h,细胞NBT染色阳性率为75%,细胞形态趋同于正常分化的细胞。同时,胞液Ca^(2+)浓度和微粒体Ca^(2+)—ATP酶活性明显降低,胞液磷酸化酶a活性则显著升高。结果提示,在DMSO作用下,HL—60细胞不仅吞噬功能增强,细胞内钙及与钙恒稳有关的酶活性也同时发生改变。 Ca^(2+) appears to have an important role in metabolism of tumor cells. We determined the variation of cytosolic Ca^(2+) concentration and microsomal Ca^(2+)-ATPase, cytosolic phosphorylase a activity during the differentiation of HL-60 cells induced by DMSO. The following results were obtained, after 120 hours culture with 1.25% (w/v) DMSO, 75% of HL-60 cells showed positive reactions of NBT reduction test. Its morphology trended to differentiated cells as well as cytosolic Ca^(2+) concentration and microsomal Ca^(2+)-ATPase activity significantly decreased while cytosolic phosphorylase a activity significantly increased. These results showed that the phagocytic functions of HL-60 cells were strengthened after the cells being induced by DMSO, cytosolic Ca^(2+) and the activity of enzymes concerned with Ca^(2+) homeostasis changed simultaneousty.
