冬凌草甲素对膀胱癌T24细胞增殖的抑制作用 被引量:5

摘要 目的探讨不同浓度的冬凌草甲素(ORI)在不同时间点对膀胱癌T24细胞增殖的抑制作用,为临床决策提供参考依据。方法将冬凌草甲素作用于T24细胞,利用酶标仪在不同时间点(λ=570nm)测定不同浓度的冬凌草甲素吸光度,同样方式用MTT法检测细胞的抑制率,流式细胞仪测定细胞凋亡率,其数值均以均数±标准差表示,每组结果由两位研究者重复测量2次校对。结果随着培养时间的延长及浓度的增加,T24细胞的吸光度数值呈明显下降趋势,差异有统计学意义(P<0.05)。浓度为8μmol/L时抑制率递增,16μmol/L时抑制率已接近50%,而32μmol/L时抑制率最高可达70%以上,变化显著,差异有统计学意义(P<0.05)。随时间、浓度梯度的变化,T24细胞凋亡率不断增加,最高可达60%以上,差异有统计学意义(P<0.05)。证明冬凌草甲素对膀胱癌T24细胞增殖的抑制具有明显的浓度-效应与时间-效应的关系。结论冬凌草甲素可诱导膀胱癌T24细胞凋亡,有明显的抗肿瘤作用。 ] Objective To investigate the inhibiting effect of Oridonin, in different concentrations and at different time points, on the proliferation of bladder cancer T24 cells, and provide reference for clinical decision making. Methods The T24 cells were affected by Oridonin. At different time points (λ=570 am) and in different Oridonin concentrations, the Oridonin absorbance was determined by microplate reader, the inhibition rate by MTI" method, and cell apoptosis rate by t]ow cylomelD. The resuhs of de, termination were expressed as mean + standard deviation. The (tetermination of each set of resuhs were repeated and proofieaded by two researchers. Results As the incubation time prologed anti concentration increased, T24 cells ahsorbance values were showed a downward trend with statistically significant difference (P 〈 0.05). The inhibition rate was steadily increased when the Oridonin concentration was 8 μmol/L, near 50% when 16μmol/L, and up to more than 70% when 32μ mol/L with statistically significant difference (P 〈 0.05). As the change of time and concentration gradient, the apoptosis rate of T24 cells was an escalation and up to more than 60% with statistically significant difference (P 〈 0.05). It was proved that the proliferation inhibition of Oridonin to bladder cancer T24 cells has significant relationship of concentration-effect and time-effect. Conclusion Oridonin can induce apoptosis of bladder cancer T24 cells and has significant anti-tumor effect.
