摘要
本文用高速离心法和DEAE-纤维素离子交换层析法从兔心肌分离提取出乳酸脱氢酶1(LDH_1)。分别以乳酸锂和丙酮酸钠为底物,测定了柠檬酸钠对LDH_1的抑制作用,结果发现,柠檬酸钠对LDH_1具有非竞争性抑制作用。结果提示,柠檬酸对糖酵解的最后一步反应可能具有一定的调节意义,并可能是巴斯德效应机理的一部分。
In this paper, lactate dehydrogenase 1 was isolated from rabbit heart muscle by the methods of supercentrifugation and DEAE-cellulose ionexchange chromatography. The inhibitory effect of citrate on this enzyme was examined when lithium lactate and sodium pyruvate were employed as the substrates respectively. The results showed that citrate expressed the noncompetitive inhibition on lactate dehydrogenase 1. The results indicated that citrate might have the regulatory effect on the last step of glycolysis and might be a part of the mechanism of Pasteur effect.
