摘要 目的摸索三种不同制片方法对乳腺疾病石蜡组织细胞的DNA进行定量分析,探讨三种方法的优劣及其在细胞DNA定量分析过程中的影响。方法本实验采用常规制片脱蜡法、常规制片脱蜡酶消法、石蜡组织制备细胞悬液法,分别对80例乳腺疾病的石蜡组织进行Feulgen染色,应用全自动DNA倍体分析仪对DNA染色玻片进行扫描分析。结果在三种方法中,常规制片脱蜡法染色结果不理想,常规制片脱蜡酶消法取得了最佳的染色效果,而石蜡组织制备细胞悬液法制备的玻片上部分区域存在一定的细胞重叠现象。结论常规制片脱蜡酶消法可用于乳腺疾病石蜡切片的DNA倍体分析。 Objective To explore the application of three different method in quantitative analysis of cellular DNA of breast tissues with disease which saved by paraffin-embedded, We analyze the pros and cons of three methods in DNA quantitative process. Methods This experiment used conventional dewaxing method, conventional dewaxing pepsin digestion method and paraffin tissue preparation of cell suspension method, 80 patients with breast disease of paraffin-embedded tissues which were carried out feulgen staining, then scan analysis of DNA ploidy were applied to stained slides. Results The result of conventional dewaxing method is not satisfactory, and the the best dyeing effect were appeared in conventional dewaxing pepsin digestion method, unfortunately, there appeared to be cell overlaps on slides which stained by paraffin tissue preparation of cell suspension method. Conclusion Conventional dewaxing pepsin digestion method can be used to analysis DNA ploidy of breast disease paraffin-embedded tissue.
