SphK1突变抑制高糖培养的系膜细胞纤维连接蛋白过度表达的研究

摘要 目的研究鞘氨醇激酶-1(SphK1)突变是否抑制了高糖培养的系膜细胞中纤维连接蛋白(FN)的过度表达。方法采用Western blot检测FN的表达。结果系膜细胞转染了SphK1突变质粒后可消除高糖诱导的FN上调效应。结论本研究结果证明了SphK1在高糖诱导的系膜细胞FN表达过程中发挥着重要的调节作用。 Objective To investigate whether SphKl depletion is responsible for the elevated FN expression in diabetic nephropathy. Methods The expression of FN were examined by Western blot. Results Transfection of SphK1 mutant abolished high glucose-induced FN expression. Conclusion These results demonstrated that the SphK1 plays a critical role in FN expression in GMC under high glucose condition.
