摘要
从威海近海采得的藻类植物经过破碎和分离筛选,得到1株红色素的高产海洋细菌,编号为SS04。经基因克隆得到16S rRNA基因序列,16S rRNA基因序列测定和分析结果显示该菌株为粘质沙雷氏菌(Serratia marcescens)。对该菌株进行发酵条件的优化探究。结果表明,海洋细菌SS04产胞内红色素的最佳碳源、氮源分别为蛋白胨和酵母粉,培养基初始pH值为7.65,胞内红色素积累最佳时间为14h,最适温度、转速分别为28℃和110r/min。
After collecting the algae plant from Weihai offshore, a marine bacteria was got which could product red pigment highly by crushing and screening separation, and then numbered SS04. The 16S rRNA gene sequence was got by gene cloning. 16S rRNA gene sequence determination and analysis results showed that the strain was Serratia marcescens. Its fermentation conditions were optimized. The results showed that the best carbon source, nitrogen source of the marine bacteria SS04 producting intracellular red pigment were peptone and yeast powder;medium's initial pH value 7.65; intracellular accumulation of red pigment best time for 14h, optimum temperature, speed were 28℃ and 110r/min respectively.
