白藜芦醇对高脂喂养C57BL/6J小鼠肝脏脂代谢的影响 被引量:1

摘要 目的探讨白藜芦醇对高脂饲料喂养的C57BL/6J小鼠肝脏脂代谢的影响。方法雄性C57BL/6J小鼠30只,按体重分为对照组、高脂组和干预组,每组10只,对照组和高脂组分别喂饲基础饲料和高脂饲料并给予0.5%羧甲基纤维素钠溶液灌胃,干预组在喂饲高脂饲料的同时给予0.5%羧甲基纤维素钠溶液溶解的白藜芦醇灌胃,第16周解剖分离小鼠肝脏,制作HE切片,测定肝脏甘油三酯(TG)、总胆固醇(TC)及游离脂肪酸(FFA)含量。结果肝脏HE切片可见高脂组肝细胞有大量脂肪空泡形成,而干预组与对照组相似,肝细胞未见脂肪变性改变;肝脏TG和TC含量,高脂组(TG为5.78±1.69 mmol/L,TC为2.69±1.20 mmol/L)均明显高于对照组(TG为4.22±1.41 mmol/L,TC为0.96±0.35 mmol/L)和干预组(TG为4.63±1.70 mmol/L,TC为1.65±0.89 mmol/L)(P<0.05),而干预组和对照组相比,差异均无显著性意义(P>0.05);肝脏FFA含量,各组间差异无显著性意义(P>0.05)。结论白藜芦醇可降低高脂喂养C57BL/6J小鼠肝脏TG、TC含量。 Objective To explore tile effect of resveratrol on liver lipid metabolism in high - fat diet C57BL/6J mice, Methods The male C57BL/6J mice (n = 30) were randomly divided into 3 groups ( 10 miee in each group) ,ineluding normal control grot,p (NC) , high fat diet group (HFD) and resveratrol intervention group (RI). NC and HFD were fed a stock rodent diet and a high fat diet respectively, and meanwhile, both were given 0.5% CMC intragastrie administration. R1 was ted a high fat diet and given resveratrol intragastric administration. After 16 weeks, separated mice livers and made HE slices, determined liver triglyceride (TG) , total cholesterol (TC) and free fatty acid (FFA). Results Observed from the liver HE slices, there were many fat empty bubbles in the HFD. R1 was the same as NC and no steatosis changes. Liver TG ( 5.78 ±1.69 ) mmol/L and TC ( 2.69 ± 1.20) ,nmoL/L content in HFD were both higher than RI ( TG :4.22±1. 41 mmol/L,TC :0.96 ±0.35 mmol/L) and NC ( TG :4.63 ±1.70 mmol/L,TC: 1.65 ±0.89 mmol/L) ( P 〈 0.05 ) , but there were not significantly different between RI and NC (P 〉0.05). Liver FFA content was not significantly different among 3 groups (P〉 0.05 ). Conclusion Resveratrol can reduce liver TG and TC content in high- fat diet C57BL/6J mice.
