摘要
目的 检测垂体腺瘤组织及原代培养的垂体腺瘤细胞中 IL- 8的表达水平 ,探讨 IL- 8与垂体腺瘤的发生和发展的相关性 .方法 免疫组织化学法检测 32例垂体腺瘤标本中IL- 8的表达水平 .酶消化法对手术切取的垂体腺瘤组织作原代培养 ,收集细胞培养上清 ,双抗体夹心 EL ISA法测定 IL - 8含量 .对组织学染色进行图像分析 ,结合肿瘤的侵袭性、病理分级等进行统计学检验 .结果 所有垂体腺瘤标本均为 IL - 8阳性 ,侵袭型垂体腺瘤的 IL- 8阳性细胞明显多于非侵袭型垂体腺瘤 (P<0 .0 5 ) ,IL - 8阳性染色强度也明显高于非侵袭型垂体腺瘤 (P<0 .0 1) .所有原代培养的垂体腺瘤细胞均有IL- 8分泌 ,侵袭型垂体腺瘤细胞上清液中 IL- 8的含量明显高于非侵袭型垂体腺瘤 (P<0 .0 1) . IL - 8的表达和分泌水平与患者性别、年龄以及分级无明显相关性 .结论 IL- 8的表达和分泌可能与垂体腺瘤的侵袭性有关 。
AIM To detect the expression of IL 8 in the pituitary adenoma tissue by immunohistochemistry and in primarily cultured cells by ELISA, and to research into the relationship between IL 8 and pituitary adenoma. METHODS Expression of IL 8 in 32 pieces of pituitary adenoma specimens was examined by immunohistochemistry. The pituitary adenoma tissue which was obtained by operation was primarily cultured and the liquid supernatant of cultured cells was collected to perform the examination for the expression of IL 8 by ELISA. The immunohistochemical staining was analyzed by image analysis system, and its outcome was statistically analyzed combined with the invasiveness and pathological classification of pituitary adenoma. RESULTS All the pituitary adenoma tissues expressed IL 8, but the number of IL 8 positive cells in the invasive adenoma was evidently larger than that of non invasive adenoma ( P <0.01). All the cells cultured from the adenoma secreted IL 8, but there was more IL 8 in the liquid supernatant of invasive adenoma cells than in that of non invasive adenoma cells ( P <0.01). The expression and secretion of IL 8 showed no close relationship with age, sex, and pathological classification of patients. CONCLUSION The expression and secretion of IL 8 may be involved in the invasion process of pituitary adenoma, so it can be used as a indicator to detect the invasiveness of pituitary adenoma.
