摘要
目的 研究胰岛素对大鼠全脑缺血再灌注损伤的治疗时间窗 .方法 造成 42只大鼠全脑缺血后 ,于再灌注后 0 ,30 min,1,3,6 ,9和 12 h等不同时间点脑室内注射胰岛素 1U,再灌注 7d后取脑进行病理检查 ,比较不同处理对大鼠海马 CA1区细胞的影响 .结果 再灌注 0 min时给予胰岛素的保护作用最为强烈 ,随着给予胰岛素时间的推迟 ,胰岛素对海马 CA1区细胞的保护作用逐渐减弱 .再灌注 6 h给予胰岛素海马 CA1区正常神经元计数为 (86± 8) mm- 1 ,是正常对照组的 48.0 % .再灌注 9h后给予胰岛素 ,海马 CA1区神经元大部分已坏死 ,与缺血对照组无显著性差异 (P>0 .0 5 ) .结论 胰岛素治疗大鼠全脑缺血再灌注损伤的治疗时间窗为 6
AIM To study the therapeutic window of insulin in the treatment of cerebral ischemic injury in rats. METHODS After the cerebral ischemia was produced in 42 rats in experimental methods, 1 U insulin was administered into the lateral ventricle at 0, 30 min, 1, 3, 6, 9, 12 h after reperfusion. Seven days later, the rat's brain tissue was taken out and examined pathologically to compare the effect of different treatments on neurons in rat's hippocampus CA1 region. RESULTS The protective effect was the most significant wheninsulin was administrated immediately after reperfusion. With the delay of administration, the protective effect abated. By administrating insulin 6 hours after reperfusion, the number of normal neurons was (86±8) mm -1 , which was 48.0% of the normal control. By administrating insulin 9 hours after reperfusion, the majority of CA1 neurons died, which had an insignificant difference from the group without treatment( P >0.05). CONCLUSION Treatment of cerebral ischemic injury in rats with insulin should be conducted within 6 hours after reperfusion.
