摘要
目的 探讨抑癌基因、细胞周期调控与胶质瘤发生、发展的关系 .方法 采用免疫组织化学技术对 6 3例脑胶质瘤组织及 10例正常脑组织标本中 p2 1WAF1 /CIP1与 p5 3基因的表达状况进行检测 ,并进行相关性分析 .结果 110例正常脑组织均为 p2 1WAF 1 /CIP1阳性表达 ,6 3例脑胶质瘤组织中p2 1WAF1 /CIP1的表达阳性率与肿瘤分化程度呈正相关 ;2 10例正常脑组织中仅 1例为突变型 p5 3弱阳性表达 .脑胶质瘤组织中突变型 p5 3的阳性率与分化程度呈负相关 ;3大部分突变型 p5 3表达阳性标本中 p2 1WAF 1 /CIP1为阴性表达 ,而大部分突变型 p5 3表达阴性的标本中 p2 1WAF 1 /CIP1 为阳性表达 .结论 p2 1WAF1 /CIP1的突变或缺失可能与胶质瘤的发生、发展有关 ,p2 1WAF1 /CIP1表达的减弱或消失可能是由 p5 3突变引起 ,野生型p5 3基因的肿瘤抑制作用可能是通过 p2 1WAF 1 /CIP1
AIM To investigate the correlation between the occurrence and development of glioma and the expression of suppressor gene and control of cell cycle. METHODS Theexpressions of p21 WAF1/CIP gene and p53 gene were observed with immunohistochemical method in 63 human glioma tissues and 10 normal brain tissues. RESULTS ① All of the normal brain tissues registered p21 WAF1/CIP1 positive expressions; the positive rates of p21 WAF1/CIP1 expression and the degrees of differentiation in glioma tissues were positively correlated. ② Only one of the ten normal brain tissues had a mutant p53 gene weak positive expression. The positive rates of mutant p53 gene expression and the degrees of differentiation in glioma tissues were negatively correlated. ③ In most of mutant p53 protein negative tissue, p21 WAF1/CIP protein was present and vice versa. CONCLUSION The loss or mutation of suppressor gene p21 WAF1/CIP may be related to the occurrence and development of glioma. The mutant p53 can cause reduction or disappearance of p21 WAF1/CIP and the expression of p21 WAF1/CIP is mainly regulated by p53. The inhibition of tumor growth by wild p53 gene may be achieved by p21 WAF1/CIP gene expression.
