摘要 目的:研究Diaphanous相关成蛋白(DRFs)家族中FMNL3基因在结直肠癌及其癌旁正常组织中的表达情况并分析FMNL3与结直肠癌发生、发展和浸润、转移的关系。方法:(1)采用免疫组化法检测240例石蜡组织标本(其中结直肠癌原发灶组织100例、淋巴结转移灶组织60例、结直肠腺瘤组织50例、癌旁正常组织30例)FMNL3蛋白的表达;(2)采用RT-qPCR技术分析FMNL3在60对新鲜结直肠癌组织及对应的癌旁正常组织中的表达情况。结果:(1)FMNL3蛋白在结直肠癌原发灶组织中的表达高于对应癌旁正常组织(阳性表达百分比分别为80%和26.7%,P﹤0.01);在淋巴结转移灶组织中的表达高于其原发灶(阳性表达百分比分别为90%和80%,P﹤0.05;其中强阳性表达百分比分别为25%和5%,P﹤0.01);在伴转移的结直肠癌组织中的表达高于不伴转移者(阳性表达百分比为86.7%和80%,P﹤0.05;其中强阳性表达百分比分别为8.3%和0,P﹤0.01))。(2)FMNL3 mRNA表达与癌细胞的分化程度、浸润深度及是否发生转移有关,癌组织低分化者共表达水平高于高、中分化者,侵犯肌层乃至浆膜层以外者其表达水平高于早期浸润者(黏膜下层以内);伴发转移的癌组织其表达水平高于未发生转移者(前者为后者的2.3倍)。结论:FMNL3与结直肠癌的发生、发展及侵袭、转移相关,并可能对其起促进作用。 Objective To explore the relationship between the expression of FMNL3,which is a member of Diaphanous-related formins family,and the development,invasion and metastasis of colorectal carcinoma by detecting the expression of FMNL3 in colorectal carcinoma and its adjacent normal tissue.Methods(1)Immunohistochemistry was used to detect the expression of FMNL3 in 240 cases of paraffin wax specimens including 100 cases of colorectal carcinoma,60 cases of metastatic tumor,50 cases of adenoma and 30 cases of adjacent normal mucosa tissure.(2)Real-Time PCR was used to detect FMNL3 mRNA expression in 60 cases of fresh specimen of colorectal carcinoma and corresponding normal tissue.Results(1)The positive detection rate of FMNL3 protein expression was higher in colorectal carcinoma than that in normal tissue(P〈0.01),higher in lymphatic metastatic carcinoma than that in primary carcinoma tissue(P〈0.05),and was also higher in primary carcinoma companied with lymphatic metastasis than that without lymphatic metastasis(P〈0.05).(2)The expression of FMNL3 was related to the invasion and metastasis potential of colorectal carcinoma although which was uncorrelate with the differentiation of carcinoma cells.In other words,the expression of FMNL3 in advanced cancer(beyond muscular layer)was higher than that in early infiltrated carcinoma(within strata submucosum),as well as the higher expression in carcinoma with lymphatic metastasis than that without.Conclusion FMNL3 may promote the development,invasion and metastasis of colorectal carcinoma.
