摘要
目的探讨不同周龄SD雄性大鼠对2型糖尿病建模成功率的影响,并分析SD雄性大鼠2型糖尿病建模的最佳时间。方法 6周龄和8周龄的SD雄性大鼠两组,普通饲料喂养,分别同时进行为期6周的高糖高脂饲养,给予30mg/kg剂量的链脲佐菌素于腹腔注射,3d后观察建模成功率(成模率);未成模的SD大鼠第2次注射同剂量的链脲佐菌素。结果 6周龄组的SD大鼠建模成功率(65.5%)比8周龄组(93.1%)低,血糖比较差异有统计学意义(P<0.01),第2次注射后6周龄组SD大鼠建模成功率(29.4%)比8周龄组(83.3%)低。结论 8周龄的SD雄性大鼠高糖高脂饲料饲养6周再结合30mg/kg剂量的链脲佐菌素是成功的2型糖尿病大鼠模型;SD雄性大鼠周龄是影响建模成功的重要因素之一。
Objective To explore the effect of different ages on the success rate of Sprague Daw ley (SD) male rat model of type 2 diabetes, and to analyze the best time for developing SD male rat model of type 2 diabetes. Methods The 6-week-old male SD rats (group A) and the 8-week-old male SD rats (group B) were selected to estab- lish SD male rat models of type 2 diabetes. The rats were fed with high-fat diet for 6 weeks, and then injected intraperi- toneally with STZ (30 mg/kg). After 3 days, the success rate of establishing model was observed. Unsuccessful model rats were injected with the same dose of STZ. Results The success rate (65.5%) of group A was significantly lower than that of group B (93.1%), with statistically significant difference in blood sugar between two groups (P〈0.01). The success rate of the 24 injection was significantly lower in group A (29.4%) than group B (83.3%). Conclusion SD male rat models of type 2 diabetes can be established successfully by feeding the 8-week-old male SD rats with high fat and sugar diet for 6 weeks and injecting 30 mg/kg dose streptozotocin. The age is one of the important factors that affect the success rate of establishing SD male rat models of type 2 diabetes.
