摘要
目的运用无血清悬浮培养体系从结肠癌细胞株COLO201中分离微球体,并证实其癌干细胞特性。方法使用无血清培养基DMEM/F12添加40ng/ml EGF、20ng/ml bFGF、B27的方法从结肠癌细胞株COLO201中分离微球体,并从长期增殖能力、克隆形成能力、长期分化能力、对放化疗的抵抗能力、迁移侵袭能力等方面证实这些微球体有癌干细胞的特性。结果从培养的第3d开始分离出微球体,微球体细胞比非微球体细胞有更强的增殖能力、克隆形成能力、长期分化能力、对放化疗的抵抗能力和迁移侵袭能力。结论使用无血清悬浮培养可以从结肠癌细胞株中分离出癌干细胞样细胞。
Objective To isolate sphere and demonstrate its cancer stem cell characteristics. Methods Serum-free medium supplemented with EGF,bFGF and B27 was used to isolate sphere from colon cancer cell line COLO201 and its cancer stem-Like cell characteristics related to proliferation, colony formation, differentia- tion, chemotherapy and radiotherapy resistance, migration and invasion ability were demonstrated. Results From 3rd day on ,the sphere was formed and sphere cells pocessed stronger proliferation, colony formation, differentia- tion, chemotherapy and radiotherapy resistance, migration and invasion ability than non-shere cells. Conclusion Spheres can be isolated from COL0201 and possess cancer stem cell-like characteristics.
