摘要
目的研究小剂量阿司匹林对糖尿病大鼠主动脉凋亡相关蛋白的作用。方法 雄性SD大鼠,尾静脉注射链脲佐菌素30 mg/kg,造成I型糖尿病动物模型,随机分为糖尿病模型组和9 mg/kg阿司匹林治疗组(ASA治疗组),每组8只,另选8只健康雄性大鼠为正常对照组。各组大鼠灌服给药12 w后,HE染色法检查主动脉组织病理显微结构,免疫组化法检测分析Caspase-3、Bax和Bcl-2的表达情况。结果 与正常对照组比较,糖尿病模型组大鼠,主动脉肌间纤维局部排列紊乱扭曲,主动脉平滑肌细胞Caspase-3、Bax表达增强,Bcl-2表达减弱,Bcl-2/Bax比值降低;ASA治疗组大鼠上述指标均有改善。结论 小剂量阿司匹林对糖尿病大鼠主动脉平滑肌细胞凋亡相关蛋白表达有调节作用。
Objective To investigate the effect of low dose aspirin on aortic apoptosis related protein in diabetic rats. Methods Male SD rats were injected streptozotocin 30mg/kg through the tail vein for establishing the type I diabetes ani- mal model, and the rats were randomly divided into diabetes model group and 9 mg/kg aspirin treatment group ( ASA treat- ment group) , 8 rats in each group. Other 8 healthy male rats were included into the control group. All the rats were given medicine. After 12 weeks, aortic tissue pathological microstructure was inspected by HE staining method, Caspase- 3, Bax and Bcl -2 expression were detected by immunohistochemical method. Results Compared with the control group, the diabetes model rats manifested such symptoms as aortic myenteric fiber turned out arrangement disorder and distortion, aor- tic smooth muscle cells Caspase -3 and Bax expression were enhanced, Bcl -2 expression was reduced, Bcl -2/Bax ratio was decreased ; as for the ASA treatment group rats, all the above indexes were improved. Conclusion Low dose aspirin might regulate the expression of aortic smooth muscle cells apoptosis related protein in diabetic rats.
