罗仙子提取物对小鼠脾淋巴细胞的体外增殖及CD4^+/CD8^+比例的影响 被引量:7

Effects of housefly maggots extracts on the proliferation and the CD4^+/CD8^+ ratio of mouser splenic lymphocytes in vitro

摘要 目的研究罗仙子提取物(housefly maggots extracts,HME)对正常小鼠脾淋巴细胞的体外增殖,以及对小鼠淋巴细胞亚群CD4+/CD8+比例的影响。方法不同浓度的罗仙子提取物对刀豆蛋白A(concanavalin A,ConA)体外处理的小鼠脾T淋巴细胞作用,初步确定其发挥作用的最佳浓度。采用MTT法检测罗仙子提取物在不同的时间点对ConA诱导的小鼠脾淋巴细胞的增殖能力,运用流式细胞仪检测细胞周期的分布以及CD4+/CD8+比例。结果罗仙子提取物浓度为40μg/mL,作用48h后,能明显促进ConA诱导的小鼠脾淋巴细胞增殖,并且能降低G0/G1期细胞的百分比以及升高S期和G2/M期细胞的百分比,并以48 h效果最明显。能使CD4+、CD4+/CD8+比例降低。结论罗仙子可能通过免疫调节作用影响动脉粥样硬化。 Objective To study the effects of housefly maggots extracts (HME) on the proliferation and the CIM +/CD8 + ration of mouse splenic lymphocytes in vitro. Methods Mouse splenic lymphocytes stimulated by concanavalin A (ConA) were treated with different concentrations of HME. The effects of splenocyte proliferation were assessed by MTF in order to select the most effective concentration of HME. Using the most effective concentration, splenic lymphocytes were treated with HME at different time points. Splenocyte cell cycle distribution and the CD4 +/CD8 + ratio were observed by flow cytometry. Results HME of 40 μg/mL notably promoted the proliferation of splenic lymphocyte after cultivation for 48 hours, reduced the G0/G1 phase cells and raised the S and GJM phase cells. HME could also reduced the CD4+ , CD8 + and CD4+/CD8 + ratio. Conclusion HME regulates immue functions in atherosclerosis.
