过氧化物酶体增殖物激活受体γ抑制高糖诱导的肾小管上皮细胞转分化 被引量:2

摘要 目的观察过氧化物酶体增殖物激活受体γ(PPARγ)在高糖诱导的肾小管上皮细胞转分化中的作用。方法大鼠肾小管上皮细胞随机分成3组:对照组(C组)、高糖组(D组)和PPARγ激动剂组(R组)。C组加正常培养液,D组在培养液中加30mmol/L葡萄糖,R组在含30mmol/L葡萄糖的培养液中加入20μmol/L的罗格列酮。在处理后6、12、24、48、72 h时用免疫细胞化学、Western blot检测上皮细胞标志物E-钙黏素(E-cadher-in)和肌成纤维细胞标志物α-平滑肌肌动蛋白(α-SMA)及PPARγ的表达。结果 C组各时间段均无α-SMA的阳性表达,而E-cadherin和PPARγ呈强阳性表达;D组12 h时出现α-SMA阳性表达,48 h达高峰并持续至72 h,而E-cadherin和PPARγ在6 h出现下降,并持续至72 h;R组各时间段均只有少数α-SMA阳性细胞,与D组比较,PPARγ在6 h出现升高,并持续至72 h。结论上调PPARγ可抑制高糖诱导大鼠肾小管上皮细胞转分化。
