高同型半胱氨酸血症大鼠主动脉组织中Bcl-2和Bax基因的表达 被引量:11

摘要 目的观察蛋氨酸灌胃诱导的高同型半胱氨酸血症(HHcy)Wistar大鼠主动脉组织中Bcl-2和Bax表达的改变。方法 20只Wistar大鼠随机分为HHcy组[予DL-重氨酸1 g/(kg.d)灌胃26周处死]和对照组(予同剂量饮用水),血清做同型半胱氨酸(Hcy)浓度测定;胸主动脉切片HE染色检测主动脉组织Bcl-2、Bax免疫组织化表达;实时荧光定量PCR法检测主动脉组织Bcl-2、Bax mRNA表达。结果 HHcy组Hcy浓度明显升高(P<0.01);HHcy组主动脉组织动脉粥样硬化斑块形成;HHcy组较对照组Bcl-2蛋白表达免疫反应积分升高,Bax蛋白表达免疫反应积分降低(P<0.05);HHcy组较对照组Bcl-2 mRNA升高,Bax mRNA降低(P<0.05)。结论蛋氨酸灌胃诱导的HHcy大鼠通过改变主动脉组织Bcl-2、Bax基因表达,从而影响大鼠主动脉粥样硬化的进程。 Objective To investigate the expression of Bcl - 2 and Bax in aorta of rats with methionine - induced hyperhomocysteinemia (HHcy). Methods Twenty Wistar rats were randomly divided into control group and HHcy group [1 g/(kg · d)]. Serum Hcy concentration was determination. HE staining was performed on thoracic aorta tissue for pathological observation, as immunohistochemistric on Bcl -2 and Bax observation was carried out. Meanwhile, real - time quantitative PCR (qPCR) assay was applied in rat aorta for assessment of Bcl -2 and Bax mRNA. Results Hcy concentration was significantly increased in HHcy group (P 〈 0. 01 ). Atherosclerotic plaque formation was observed in aorta in HHcy group. Significant up - regulation in Bcl - 2 and clown - regulation in Bax were revealed in HHcy group according to both immunohistochemistry and qPCR ( P 〈 0. 05 ). Conclusion Methionine induces HHcy by affecting the expression of Bcl - 2 and Bax. thus exacerbates the process of aortic atherosclerosis in rats.
